Molecular detection of Anaplasma platys in Costa Rican dogs

Abstract

The presence of Anaplasma platys, causal agent of infectious canine cyclic thrombocytopenia (ICCT) was analyzed in Costa Rican dogs using a polymerase chain reaction (PCR) assay. A total of 300 blood samples were analyzed; 20 samples were collected from dogs with suspected ICCT, while the other 280 were obtained from blood bank samples previously collected from dogs with suspected ehrlichial infection. The following information was collected for each sample: geographical location, period of sample collection, race, gender, and age. Blood samples were analyzed using two different PCR assays: one to amplify a fragment of the 16S rRNA gene of the Anaplasmataceae family and a nested PCR to specifically amplify A. platys. A total of 19 samples (6.3%) were positive for A. platys, showing the presence of the agent in dogs in Costa Rica. A strain of A. platys showed 100% similarity to a partial sequence of 16S rRNA gene of A. platys (AF156784) published in GenBank. Anaplasmataceae PCR detected less samples being positive for A. platys than nested PCR; however, this study showed evidence regarding the probable presence of other species of the Anaplasmataceae family in dogs in Costa Rica.
